Effect of cryoprotectant on the motility, viability, fertilization, and DNA integrity of naleh fish Barbonymus sp. (Cyprinidae) sperm

Naleh fish Barbonymus sp. is a commercial freshwater fish, which indigenous to Aceh, Indonesia. The population of this species has declined over the years as result habitat perturbations and overfishing. Hence, crucial need develop cryopreservation method support breeding programs. This involved use cryoprotectant an important component. objective study, therefore, was explore best for naleh spermatozoa, total five types were tested. These include DMSO, Methanol, Ethanol, Glycerol, Ethylene Glycol at similar concentration 10%, individually combined with 15% egg yolk, every treatment performed in three replications. Conversely, Ringer’s solution adopted extender, sperm cryopreserved liquid nitrogen 15 days. results showed significant influence on motility viability, well fertility (P <0.05), although DMSO provided outcome, compared others 47.17%, 50.13%, 45.67%, respectively. Furthermore, DNA fragmentation had not occurred fresh samples, indicating protective effect tested cryoprotectants. It concluded that 10% yolk spermatozoa.
